I'm very inexperienced and would appreciate any assistance offered. I'm trying to come up with some budgetary numbers to work out if we can afford to pursue a new build. I would like to come up with some rough costings for a build mostly at this stage so I can work out whether we should buy a block of land I've got my eye on. If the build costs are going to exceed our budget we won't buy the block and buy pre-existing instead.
I'm looking at a block of land in the bayside area of east Brisbane. The block is fairly flat and approximately 800sq mtrs. I like the look of building a Clarendon Madison City 40 with hamptons facade. It is 374 sq mtrs and a 5/2.5/2. List price from the Clarendon brochure is $323000, although that price makes no mention of which facade and I'm assuming Hamptons will be more expensive. We would also like to have a pool.
I don't think we'll be going berserk with high end fit out and landscaping etc but we would like it to look nice.
So the question is how much more than that $323000 number is the total build likely to be? Obviously we're talking rough numbers here. Is this likely to be achievable for say $500 000?
What I'm struggling to come to grips with is calculating the build cost on a $ per sq mtr basis. if I used $1500 per sq mtr x 374 sq mtrs that says it should be approx $560 000. So where is the extra $230000 over the Clarendon base price coming from.

Definitely achievable for $500,000
I got a quote on the Madison 39, which isn't that much different in price to the City 40. With a few changes and bits and bobs added, I had a quote of $393,000.
With the extras you add at Tiles/colours/electrical, depending on the finish you want you could remain fairly close to that or add a whole heap on.
Clarendon's price is VERY basic. No driveway, landscaping, window coverings, 2 coats of paint etc. So their advertised price is I guess about $880 m2 average across the designs. I've heard $1500m2 is for really quite a nice house. I'm estimating about $1150m2 for my current build, and i'm pretty stoked with what i'm getting with that.
Have a look at my blog as well as TiredHalo's - gives you an idea of the additional pricing for some things. As an example, the standard facade is included - it's quite unattractive. The next facade up costs $5000, and that's not including render onto the facade - just to get the structure of the facade. So straight away you've added $8-10k onto that base price. FYI Hampton facade is about $30k from memory.

Keep learning about the building industry so you know the process and what's coming up, you will get a better result if they know your not so inexperienced, you will be eaten alive if they think anything can be put past you, try and include everything before you sign contract, as they will shock you with their pricing once you have signed on, the more you learn, the better chance you will get a better build.

Go to a display home and talk to the reps about what level of finish the display homes have a the cost of the different facade. Some of the display homes will be up spected more than $100k from the base model. Look at what is include and work out what the extras will cost you. Most builders heavily under spec things like number of powerpoints and base price will be Batton light fittings rather than downlights etc. every builder is different so it’s hard to give exact figures.

One quick comment - it looks as though you're a bit fixated on a cost per square metre, but that's really a fairly farcical number. It may have some value in comparing two things that are identical in every other way, but between two house designs (even from the same builder with the same inclusions!) it just doesn't seem to happen.
You need to look at totals, and if trying to get an idea of cost you really can't expect to extend a house by 20% and pay 20% extra (nor shrink a house by 20% and pay 80% of the cost).
Also, what people say is easily achievable varies widely from metropolitan to regional, and between states as well. So if checking whether something can be done for $X, you need to be sure they're in the same market as you are (location & time - what we paid 5yrs ago is probably irrelevant now).
PS. if budget is tight get a quote for the pool ASAP, before you make any decisions. A mate was quoted ~$70k for a pretty simple backyard pool with easy access a couple of years ago, that's a big chunk out of a build budget.
