There are actually 4 on that plan, we'll be removing the powder room in middle.

1 in laundry, main and ensuite to stay
How big is this plan? It looks like you are trying to squeeze too much in.

I certainly wouldn't be getting rid of the pwd in lieu of the laundry toilet - who wants a toilet right off the kitchen?

The dining and family are around the wrong way IMO.

I would move the activity room to between main bathroom and bed 2.

That is tight for a 4 bed 3 living plus study plus IT nook house IMO. The kitchen is small too. Definitely continue to rejig and work out what you really need/want. If you make spaces too small it doesnt matter how many you have, if it makes them unusable .
I recon mirror the meals area and kitchen/Laundry/ Buttlers (back to old position but in this layout) the activity room is smaller than the home theater. That extra space can be given to the kitchen and make the laundry less cramped.
This could also increase your threatre space if you wanted.

You main areas are suffering due to excessive robes and storage areas. your trying to fit too much into the space, something has to give. (hopefully not your main entertainment areas)
this is the latest - some changes are in draft that i have circled - adding in storage in a couple of areas, servery window off kitchen to alfresco and main bathroom having seperate toilet. Laundry sliding door to PWDR will also open in middle sliding so that there is more bench space.

Looking at sourcing large sliding door for activity space to open/close.

The alfresco still shades too much of your living areas. Why don't you move it further to the west, over the laundry, so you allow more northern light into the living areas. Appropriate length northern eaves for the rest of the living area.
